Has anyone got advice or experience of measuring the contrast between text
superimposed a video background?

I would imagine that you have to take stills of the video, sample pixels of
the text and background, and test the colour values. But there could be
thousands of frames in, say, a 1 minute video.

Has anyone got practical tips?

I don't have any immediate recommendations for testing tools, but would
recommend using an outline around the text. A stroke that contrasts
sufficiently with the text itself should enhance contrast on a variety of
backgrounds. Alternatively, you could have a background behind the text, as
YouTube does.

I thought video was exempted from 1.4.3 and 1.4.10 (High Contrast Non-text). Is this correct?
