Jared is right. Another tip is to mute the "speakers" in the Elluminate
platform and try that. I used to be an Elluminate administrator and this was
a common "audio feedback" issue. You mentioned to get the same sound when
you recorded the audio. If this is the case, you may need to tweak the
settings in your mixer.

This post is rather off-topic, but...

It sounds like a basic feedback loop to me. The microphone is picking up the
audio, and when the presentation software opens, it plays the microphone
audio, which the microphone then picks up again and amplifies over and over
until it's just a squeal. You might be able to address this by changing
settings to ensure that your own microphone audio never plays through the
speakers. I can't give you specific directions as you have a fairly
complicated set up, but that seems to be the general issue. Or simply use
headphones rather than your speakers.
