Is there a compelling reason NOT to use the Acrobat 'Role Map' as far as
AT software is concerned? I have some really large PDFs to remediate, and
the tags generated by Acrobat's Autotag are all non-HTML5-standard. Hand
editing them, which is what I have been assiduously doing, is getting
REALLY tiresome! :)

First... a point of clarification. Role-mapping structure elements in PDF is a PDF feature. Software that supports Tagged PDF supports role-mapping; not just Adobe.
PDF's role-mapping feature is expressly intended to allow users to create their own tags for their own purposes while also ensuring the tags are usable by AT (or other re-use software).
So.. to answer the question: role-map away! The feature exists precisely to solve problems like the one you've got.
