Hello everyone,
I have a dropdown where let's say the dropdown list is Male and Female and
along with Please choose a gender is the default value within the dropdown.
When a screen reader user navigates to it, the screen reader announces
Please choose a gender but upon choosing one replace the option with either
Male or Female and so the screen reader announces Female (along with role
and state) but there is no visible label available. Does this directly fail
any WCAG/Section 508 guideline(s)?

Yes, it fails SC 3.3.2: Labels or Instructions, because there must be a persistent visible label.
It may also fail SC 4.1.2: Name, Role and Value, but I can't tell without seeing the code. You can't make that decision based on assistive technology behaviour.
