WAI-ARIA includes roles for keyboard-accessible Tooltips, Definitions, and Dialog Tooltips. Below are some good resources related to these ARIA roles.

WAI-ARIA 1.0 | The Roles Model:

Definition: http://www.w3.org/WAI/PF/aria/roles#definition

WAI-ARIA Authoring Practices 1.0 | Design Patterns:

Dialog Tooltip (in progress): http://www.w3.org/WAI/PF/aria-practices/#dialog_tooltip

DHTML Style Guide:

Dialog (Tooltip): http://dev.aol.com/dhtml_style_guide#dialogtooltip

Working code examples (recommend using with IE8/Firefox 3.6 and JAWS 11, or higher):

Subject: [WebAIM] Interactive Glossary

I've been tasked with a project that includes a request for an
"Interactive Glossary" that would provide a word's definition in a
pop-up window when users hover over a word. The voices in my head tell
me there's not a way to do that and make the definitions accessible. Am
I wrong? Can anyone provide examples or advice on how to create
something like this?
