I'd use both ANDI and manually testing with a screen reader like NVDA.
With ANDI I'd use Focusable Elements and navigate through all items to see what it reveals. Then I'd use the graphics/images tool then the links/buttons tool, structures, the tables tools (if any were present), and test iframes if those were present. Each of the ANDI components is best utilized with the huge/long PDF they have that has you check against the WCAG success criteria, which I believe is still only using the 2.0 AA level.
Having gone through the Trusted Tester certification process I value what the ANDI tool can do but like Steve said, you'd still need to try out things with a screen reader.
